RNA Synthesis, Fluorescence-based Probe Design and Characterization

The aim of this project is the design, development, characterization and, on larger scale, generation of novel fluorescent nucleotide base analogues that can be used to label si/miRNA, ASOs (antisense oligonucleotides) and mRNA. Such fluorescent molecules will be further incorporated into shorter oligonucleotides using solid-phase synthesis as well as into longer sequences using click-chemistry and/or enzyme based technologies and spectroscopically characterized for further use in advanced single-cell microscopy studies. The so generated oligonucleotide complexes will facilitate characterization of cargo heterogeneity, cellular uptake, trafficking and release of nucleic acids delivered using LNPs, EVs and exosomes as vehicles.
